This product is probably about as close as you can get to the niagen supplements, like Thorne ResveraCel, without actually containing niagen or using the patented process. Nicotinamide is a form of niacin easily processed by the body. And it contains bonus resveratrol. I've only been taking these two weeks, but I've found these supplements give me some mental boost/clarity and physical energy. My skin seems more elastic. I have less appetite (my usual tendency is to over eat) and sleep better. I've lost a couple of pounds. One side effect I get is upset stomach, but the seller/manufacturer claims this will go away with time. Overall, while these supplements are no "miracle cure," I think they are providing some benefit, and I will continue to take them. I'll reevaluate once I've made it through the first bottle.

What Does NMN Supplement Do?

Chromadex NMN Supplement Test Report: 64% of NMN Supplements Fail Tests

ChromaDex NMN Report (Oct 2021): Supplement manufacturer ChromaDex alleges that testing has revealed that more than half of NMN anti aging supplements being sold on Amazon contained almost none of the bioactive molecule.

ChromaDex markets TruNiagen, which is based on its branded Niagen ingredient, which is a form of nicotinamide riboside (NR), the form of nicotinamide that actively crosses into the cell to participate in the NAD+ pathway. Boosting this pathway, and thus maintaining cellular function at a lively, youthful level, has become a prominent theme in recent anti aging research.

Some researchers have argued that a transporter exists for NMN too, which is something that ChromaDex disputes. It claims that NMN must first be converted into NR in the blood before it can be used by the cells.

In the several years since the NMN transporter discovery was asserted, the NAD+ anti aging field has become crowded with NMN based supplements all seeking a piece of the pie created by ChromaDex and its competitor and one time customer Elysium. (The two companies have been locked in various patent infringement and breach of contract suits for several years.)

Last fall, ChromaDex obtained 22 samples of NMN products being sold on Amazon. Of these, 14 claimed to contain 500 mg of NMN. A further two products claimed 300 mg on the label; five claimed 250 mg and one product claimed 125 mg.

Only one of the 500 mg products came close to meeting label claim. It had 456 mg of NMN. All of the other 13 products in the 500 mg group either came in at below reporting limit (BRL, defined as less than 1% of the stated amount) or, in the case of 3 products, no NMN was detected at all.

Almost all of the rest of the products performed better, having at least 88% of the label claim. A lone 250 mg product was identified as BRL.

In summary, ChromaDex said that 64% of the products tested contained less than 1% of the stated amount of the active ingredient, which should give consumers pause, the company said.

"While this is a limited snapshot of the vast NMN finished product landscape, it does provide a glimpse into the high variability of product quality that is available... According to this study, the majority of the products one might purchase online contain such a small amount of NMN that there would be no clinical benefits achieved from the dose. Another concern with these adulterated products is that the actual contents are not known and could pose a risk to the user," the company said in a statement.

Editor's Note: In February 2022, another third-party investigation uncovered tens of thousands of fake supplements sold on Amazon. Many of these included NMN.

David Sinclair Supplements List

Keep in mind that to date, Dr. David Sinclair has made a point to not promote or endorse any supplement products. We compiled this list from interviews and books in which Professor David Sinclair mentions supplements he takes.

We don’t know if he still takes these supplements, or whether he takes additional supplements that are not included on this list.

For example, in his book Lifespan, he mentions on page 304 that he takes NMN and resveratrol every morning, often mixed in his yogurt.

He is also a co-founder of a pharmaceutical company called MetroBiotech which has begun clinical trials with a special, crystalline form of NMN called MIB-626.

But this will presumably be for sale as a drug to treat specific medical conditions, if the studies show positive outcomes.

NR vs NMN Molecule

Pitting NR vs NMN against each other is, for now, somewhat of a moot point because the two molecules have never been studied side by side in humans. The biggest, and most obvious, difference between NMN and NR is size. NMN is simply larger than NR, meaning it often needs to be broken down to fit into the cell. NR (Nicotinamide Riboside), when compared to other NAD+ precursors (like nicotinic acid or nicotinamide) reigns supreme in efficiency.

But give NMN a new door, one it can fit through, and it’s a whole new game. This is where cellular transporters come into play. Transporters are proteins that are doors on the cell; they allow molecules to enter the cell without needing to chemically transform.

Research from Shin-ichiro Imai, M.D., Ph.D., a professor of developmental biology at Washington University in St. Louis, identified a transporter that allows NMN to get into the cell without converting to NR. The catch? The transporter is only on cells located in the gut of mice and only works in the presence of sodium ions. NR, however, has been shown to enter cells in the liver, muscle, and brain tissue of mouse models. To date there is not yet evidence that these mouse studies can be extrapolated to humans. But again, the two have never been matched up against each other in a way that can truly identify one as superior to the other.

As research grows, it is likely to become clearer that NMN and NR may have differing benefits depending on the part of the body and their use. Where NMN transporters are found, they could make one more preferable over the other. NMN has also been found to improve insulin activity and production which could help to accelerate metabolisms and make the body more glucose tolerant.

Stability and storage of NMN and NR

A stability analysis demonstrates NR is stable (does not undergo degradation) for up to six hours at room temperature and seven days at two to eight degrees Celsius. Regarding stability of NMN and NR, David Sinclair, PhD, A.O. of Harvard University says, “Make sure your NR and NMN is kept in the cold. If it’s just on the shelf and it’s not in a stabilized form, then it will degrade to nicotinamide, which is something you don’t want to take high doses of because we’ve shown in my lab many years ago that nicotinamide will inhibit the sirtuins, and PARP as well, and interfere with DNA repair.”

What Foods Contain NMN?

The aim of taking NMN supplements is to boost NAD levels. So you need to look out for foods that are rich in NAD precursors, not just NMN. As lots of different molecules can be converted to NMN, and then NAD.

• Vitamin B3 rich foods: Foods that are high in niacin (vitamin B3) can help increase NAD+ levels in the body. Good sources of niacin include yeast, beef liver, poultry, fish, and whole grains.

• Leafy greens: Leafy greens, such as spinach and kale, are rich in folate, which can help increase NAD+ levels in the body.

It is important to note that foods which are high in NAD precursors do not necessarily increase NAD or NMN levels. Despite having an abundance of NAD precursors in your diet, your cells can use these nutrients for other processes and may not use them to produce NMN and then NAD.

On the other hand, NMN supplements push your cells to produce more NAD which has been shown in several animal and a few human studies. 

NMN is available in vegetables (broccoli and cabbage), avocado, mushrooms, meat, and shrimp but trying to obtain sufficient through diet alone would be problematic. 

Study dosages have been about 50mg - 250mg and suggested that a 150 pound (68kg) human would require about 560mg per day. The difficulty is that you would have to eat about 100 pounds of edamame, 1,800 pounds of broccoli, or similarly impossible amounts of cucumber, cabbage, avocado, tomato, mushrooms, raw beef, or shrimp.

What makes a good NMN supplement?

With so many different supplements on the market claiming to be the best, it's hard to know which one is worth investing in. Realistically, there are only a few things that really matter:

1. Purity

The best NMN supplements use 98% purity NMN. It's as simple as that, always look for this level and nothing less should make the cut. After all, it's obvious that we all want a supplement that contains as much of the active ingredient as possible.

2. Dosing

There is no official recommended dose of NMN. It's a relatively new supplement and most brands contain between 250mg - 750mg per serving which is based on the limited amount of research on this ingredient. David Sinclair openly says that he takes 1000mg of NMN a day which is a relatively high dose.

A recent study published in February 2023 tested daily NMN at doses of 300mg, 600mg, and 900mg over a 60-day period in healthy adults of 40-65 years old.

There were no side effects reported and they found that NAD levels increased in all of these doses. It's important to highlight this was a small study of only 80 individuals and the study only ran for 60 days so it's not clear what the long-term outcomes will be. At this stage, most people think that they'll just take Vitamin B3 and that will eventually form NMN. But the issue is that a lot of energy is required to go from vitamin B3 to NMN and our cells won't naturally go down this route.

What are the side effects of taking NMN supplements?

As with all things NMN related, there isn't much solid information on NMN side effects. There are only a handful of studies using NMN in humans. Side effects are seldom reported and doses up to 1000mg are generally well-tolerated.

For example, one study gave 250 mg/day of NMN for 10 weeks to 25 prediabetic women who were overweight or obese without any adverse effects. (2)

But this does not mean that you won't experience any side effects. As with any supplement, the risk of experiencing a negative reaction is always possible. So always discuss this with your physician, especially if you have any other medical conditions and taking any medicines.
